How does the tuner tell whether a string is in tune?

It shows the detected pitch, the target string and the deviation in cents at the same time; the closer the cents reading is to zero, the closer the string is to its target.

Why does it need the microphone?

The tuner listens to the string to analyze its pitch. Microphone audio is used for real-time tuning on the current device.
